本發(fā)明涉及基因工程領(lǐng)域,具體涉及提高比活的α-淀粉酶BaAmy突變體及其編碼基因和應(yīng)用。
背景技術(shù):
:α-淀粉酶能夠能跨越不能切分支點(diǎn)的α-1,6鍵從淀粉分子內(nèi)部隨機(jī)地切開α-1,4糖苷鍵。α-淀粉酶作為一種重要的水解酶廣泛的應(yīng)用于食品加工、酒精釀造、制藥、紡織退漿、造紙、洗滌液及飼料等多種領(lǐng)域。目前工業(yè)上廣泛使用的商品化α-淀粉酶和糖化酶的最適pH值約為6.5和4.5,因此在液化和糖化過程中期間需要加入酸堿來調(diào)節(jié)pH。在液化和糖化過程中大量加入酸堿不僅使加工工藝復(fù)雜化,而且增加了生產(chǎn)成本。如果能夠開發(fā)出在酸性條件下穩(wěn)定的α-淀粉酶,則在液化和糖化過程中不需要額外添加酸堿進(jìn)行pH調(diào)節(jié),不僅可以減少試劑消耗,簡化加工工藝,而且可以降低生產(chǎn)成本,節(jié)約糧食。對淀粉加工領(lǐng)域具有重大意義。酸居芽胞桿菌α-淀粉酶簡稱BaAmy,雖然具有很好的酸穩(wěn)定性,但是其比活低,生產(chǎn)成本高,限制了其工業(yè)化應(yīng)用。因此,提高BaAmy的比酶活,降低其生產(chǎn)成本,是BaAmy工業(yè)化應(yīng)用急需解決的問題。技術(shù)實(shí)現(xiàn)要素:本發(fā)明的目的是通過對來源于酸居芽胞桿菌(Bacillusacidicola)的酸性α-淀粉酶BaAmy進(jìn)行分子改造,使改造后的酸性α-淀粉酶具有更高的比活,降低生產(chǎn)成本,為酸居芽胞桿菌酸性α-淀粉酶的工業(yè)化應(yīng)用奠定基礎(chǔ)。本發(fā)明的目的是提供提高比活的α-淀粉酶BaAmy突變體的編碼基因。酸居芽胞桿菌(Bacillusacidicola)的酸性α-淀粉酶BaAmy的核苷酸序列和氨基酸序列如SEQIDNO.1和SEQIDNO.8所示。本發(fā)明采用定點(diǎn)飽和突變的方法對SEQIDNO.8所示的α-淀粉酶BaAmy的第37位、第85位、第191位、第279位、第291位、第319位和第333位進(jìn)行分子改造,經(jīng)過高通量篩選得到一系列提高比活的α-淀粉酶突變體。這些突變體的核苷酸序列如SEQIDNO.2到SEQIDNO.7所示,氨基酸序列如SEQIDNO.9到SEQIDNO.14所示。本發(fā)明通過蛋白理性改造和高通量篩選技術(shù)對酸居芽胞桿菌(Bacillusacidicola)的酸性α-淀粉酶BaAmy進(jìn)行分子改造。相對于原始α-淀粉酶的比活,突變后α-淀粉酶比活的提高幅度為45%-110%,為酸居芽胞桿菌酸性α-淀粉酶的工業(yè)化應(yīng)用奠定基礎(chǔ)。附圖說明圖1原始α-淀粉酶和突變體BaAmy-1至BaAmy-6的最適反應(yīng)pH圖2原始α-淀粉酶和突變體BaAmy-1至BaAmy-6的pH穩(wěn)定性圖3原始α-淀粉酶和突變體BaAmy-1至BaAmy-6的最適反應(yīng)溫度圖4原始α-淀粉酶和突變體BaAmy-1至BaAmy-6的熱穩(wěn)定性具體實(shí)施方式以下實(shí)施例中未作具體說明的分子生物學(xué)實(shí)驗(yàn)方法,均參照《分子克隆實(shí)驗(yàn)指南》(第三版)J.薩姆布魯克一書中所列的具體方法進(jìn)行,或者按照試劑盒和產(chǎn)品說明書進(jìn)行;所述試劑和生物材料,如無特殊說明,均可從商業(yè)途徑獲得。實(shí)驗(yàn)材料和試劑:1、菌株與載體酸居芽胞桿菌(Bacillusacidicola)購自中國工業(yè)微生物菌種保藏管理中心,菌種編號為21144、大腸桿菌菌株Topl0、畢赤酵母X33、載體pPICzαA,Zeocin購自Invitrogen公司。2、酶與試劑盒Q5高保真Taq酶MIX購自NEB公司,質(zhì)粒提取,膠純化,限制性內(nèi)切酶、試劑盒購自上海生工公司。3、培養(yǎng)基大腸桿菌培養(yǎng)基為LB(1%蛋白胨,0.5%酵母提取物,1%NaCl,pH7.0)。LBZ為LB培養(yǎng)基加25ug/mLZeocin。酵母培養(yǎng)基為YPD(1%酵母提取物,2%蛋白胨,2%葡萄糖)。酵母篩選培養(yǎng)基為YPDZ(YPD+100mg/Lzeocin)。酵母誘導(dǎo)培養(yǎng)基BMGY(I%酵母提取物、2%蛋白胨、1.34%YNB、0.00004%Biotin、1%甘油(V/V))和BMMY(除以0.5%甲醇代替甘油,其余成份相與BMGY相同)。實(shí)施例1、酸居芽胞桿菌(Bacillusacidicola)的酸性α-淀粉酶的克隆將酸居芽胞桿菌接入LB培養(yǎng)基,培養(yǎng)24小時后,提取其基因組DNA。根據(jù)已報道酸居芽胞桿菌α-淀粉酶的序列(Genebank:JN680873)設(shè)計(jì)兩條引物(R:5'-ATGAATTCAACGGCACCATGATGCAGTAT-3'和F:5'-GCTCTAGACTAAACCGAACCACCATTGACTTTG-3')用于擴(kuò)增酸居芽胞桿菌α-淀粉酶基因。將擴(kuò)增的PCR產(chǎn)物純化回收,連接到表達(dá)載體pPICzαA,得到表達(dá)載體pPICzαA-BaAmy。實(shí)施例2、定點(diǎn)突變以上述pPICzαA-BaAmy為模板,用相應(yīng)的引物進(jìn)行PCR擴(kuò)增,具體地?cái)U(kuò)增反應(yīng)體系如下:Q5高保真Taq酶MIX23uL對應(yīng)突變體引物1uL對應(yīng)突變體引物1uLpPICzαA-BaAmy(20ng)2uL加水至50uL反應(yīng)程序如下:瓊脂糖電泳檢測PCR擴(kuò)增結(jié)果,純化回收PCR產(chǎn)物。用限制性內(nèi)切酶DpnI將原始質(zhì)粒分解,將分解完的產(chǎn)物才用熱激法轉(zhuǎn)入大腸桿菌Top10,通過菌液PCR驗(yàn)證重組轉(zhuǎn)化子,提取驗(yàn)證正確的轉(zhuǎn)化子的質(zhì)粒進(jìn)行測序,從而確定相應(yīng)的突變體。將測序正確的突變體,用SacI線性化,轉(zhuǎn)入畢赤酵母X33。實(shí)施例3、高通量篩選高比活突變菌株將實(shí)施例2中的酵母重組轉(zhuǎn)化子用牙簽逐個挑至24孔板,每個孔中加入1mL含有BMGY培養(yǎng)基,30℃,220rpm培養(yǎng)24h左右,離心去上清。再分別加入1.6mLBMMY培養(yǎng)基進(jìn)行誘導(dǎo)培養(yǎng)。培養(yǎng)24h后,離心取上清,將上述上清液分別取出200μL至96孔板,進(jìn)行α-淀粉酶酶活測定。α-淀粉酶酶活檢測參照中華人民共和國國家標(biāo)準(zhǔn)《GB/T24401-2009》進(jìn)行測定。實(shí)施例4、組合突變將實(shí)施例2中酶比活提高的單突變位點(diǎn)T37N,T37L,T37P,T37W,T37K,T85R,T85G,T85Q,T85E,T85A,N191F,N191L,N191P,N191V,N191I,S241F,S241A,S241T,S241V,S241N,N279D,N279V,N279P,N279Q,N279F,S291N,S291K,S291I,S291W,S291R,T319H,T319C,T319G,T319E,T319Y,V333G,V333P,V333N,V333A,V333D,分別進(jìn)行組合,實(shí)驗(yàn)過程同實(shí)施例2相同。通過實(shí)驗(yàn)最終得到6個組合突變分別命名為BaAmy-1、BaAmy-2、BaAmy-3、BaAmy-4、BaAmy-5、BaAmy-6其中BaAmy-1包含的突變位點(diǎn)為:T37L,T85G,N191P,S241A,N279D,S291N,T319H,V333G。其中BaAmy-2包含的突變位點(diǎn)為:T37N,T85A,N191F,S241F,N279D,S291N,T319H,V333G。其中BaAmy-3包含的突變位點(diǎn)為:T37P,T85R,N191L,S241V,N279V,S291K,T319C,V333P。其中BaAmy-4包含的突變位點(diǎn)為:T37W,T85Q,N191V,S241T,N279P,S291K,T319E,V333A。其中BaAmy-5包含的突變位點(diǎn)為:T37K,T85E,N191I,S241N,N279V,S291W,T319Y,V333P。其中BaAmy-6包含的突變位點(diǎn)為:T37W,T85Q,N191V,S241N,N279H,S291K,T319Y,V333G。實(shí)施例5、原始α-淀粉酶BaAmy及α-淀粉酶突變體的比活分析分別將原始α-淀粉酶BaAmy和突變體α-淀粉酶進(jìn)行純化,純化方法為鎳柱純化。將純化好的α-淀粉酶和突變體α-淀粉酶分別測定相應(yīng)的酶活并計(jì)算出比活。以突變體比活除以原始α-淀粉酶比活,來計(jì)算突變體比活的提高幅度。相比原始BaAmy,突變后的BaAmy比活提高幅度為45%-110%(具體結(jié)果見表1)。表1原始α-淀粉酶和突變體α-淀粉酶相對比活編號相對比活(%)原始α-淀粉酶100BaAmy-1145BaAmy-2150BaAmy-3180BaAmy-4175BaAmy-5210BaAmy-6195實(shí)施例6、原始α-淀粉酶BaAmy及α-淀粉酶突變體BaAmy-1至BaAmy-6最適反應(yīng)pH及pH穩(wěn)定性參照國標(biāo)方法測定原始α-淀粉酶BaAmy及α-淀粉酶突變體BaAmy-1至BaAmy-6的最適反應(yīng)pH。BaAmy及BaAmy-1至BaAmy-6的最適反應(yīng)pH如圖1所示。由圖1可知,突變體BaAmy-1至BaAmy-6的最適pH并沒有發(fā)生太大變化,最適反應(yīng)pH值均為4.5,幾乎和BaAmy一樣。將BaAmy及α-淀粉酶突變體BaAmy-1至BaAmy-6分別在pH4-8條件下室溫處理2小時,然后參照國標(biāo)的方法測定酶活。BaAmy及α-淀粉酶突變體BaAmy-1至BaAmy-6的pH穩(wěn)定性如圖2所示。相比BaAmy,突變體BaAmy-1和BaAmy-5具有更好的耐酸性,在pH4條件下,剩余酶活分別為95%和96%。而突變體BaAmy-2、BaAmy-3、BaAmy-4和BaAmy-6的pH穩(wěn)定性與BaAmy一致。實(shí)施例7、原始α-淀粉酶BaAmy及α-淀粉酶突變體BaAmy-1至BaAmy-6最適反應(yīng)溫度及熱穩(wěn)定性參照國標(biāo)方法測定BaAmy及突變體BaAmy-1至BaAmy-6的最適反應(yīng)溫度。BaAmy及突變體BaAmy-1至BaAmy-6的最適反應(yīng)溫度如圖3所示。由圖3可知,BaAmy及突變體BaAmy-1至BaAmy-6的最適反應(yīng)溫度均為60℃。將BaAmy及突變體BaAmy-1至BaAmy-6分別在50℃-90℃條件下水浴處理30分鐘,然后參照國標(biāo)的方法測定酶活。BaAmy及突變體BaAmy-1至BaAmy-6的熱穩(wěn)定性如圖4所示。由圖4可知,突變體BaAmy-2和BaAmy-6熱穩(wěn)定性要好于BaAmy。在90℃條件下水浴處理30分鐘后,突變體BaAmy-2和BaAmy-6的剩余酶活分別為85%和80%,而BaAmy的剩余酶活為50%。突變體BaAmy-1、BaAmy-3、BaAmy-4和
