本發(fā)明涉及對真核生物同時進行基因敲除和基因過表達。
背景技術(shù):
基因支持著生命的基本構(gòu)造和性能。儲存著生命的種族、血型、孕育、生長、凋亡等的全部信息。人類根據(jù)需要進行基因操作,選擇目的基因(dna片段)在體外構(gòu)建載體,轉(zhuǎn)移至另一細胞或生物體內(nèi),以達到改良和創(chuàng)造新的細胞或個體或治療人類疾病的目的?;虿僮鞯姆绞接泻芏喾N,主要可分為基因的過表達和降低表達或基因敲除?;蜻^表達技術(shù)已經(jīng)非常成熟,主要由一個啟動子啟動外源基因表達即可。基因敲除技術(shù)則不然。傳統(tǒng)的基因操作通過基于全能胚胎干細胞的同源重組實現(xiàn)。需要構(gòu)建一個復(fù)雜的同源重組載體,包括了陽性和陰性篩選抗性等,效率非常低,而且操作復(fù)雜、耗時長,很難用于實際應(yīng)用。
一些生物性狀受到激活基因和抑制基因的同時調(diào)節(jié)。在對這些性狀進行調(diào)控時,往往需要對影響這些性狀的激活基因和抑制基因同時進行敲除和過表達。由于傳統(tǒng)的基因敲除非常復(fù)雜,因而利用傳統(tǒng)的方式同時進行基因敲除和基因過表達非常困難。通常,為了實現(xiàn)在同一個體或同一細胞進行基因敲除和基因過表達這一目的,需要先進行基因敲除再進行基因過表達,或者是先進行基因過表達再進行基因敲除來實現(xiàn)。很顯然,這樣的策略操作步驟繁瑣、效率低、耗時長。要進行多基因的敲除或過表達,則更是難上加難。特別是新分離的原代細胞,因為生長時間短,基因?qū)肜щy,不能先進行基因敲除或過表達,再進行過表達或基因敲除,必須同時進行基因敲除和基因過表達。這樣傳統(tǒng)的策略幾乎是不可能的。
近年來出現(xiàn)了一類新的基因敲除技術(shù),這類技術(shù)利用不同的機制,將核酸內(nèi)切酶導(dǎo)向到基因組的特定位點,核酸內(nèi)切酶將dna剪切,造成dna雙鏈斷裂,實現(xiàn)dna序列改變。為區(qū)別于80年代發(fā)明的基因打靶技術(shù),這類新的技術(shù)被稱之為基因編輯技術(shù)(geneediting)。這類技術(shù)主要包括zfn、talen和crispr/cas9。crispr/cas9是2012年人們研發(fā)出的一種新型基因組修飾技術(shù)--規(guī)律成簇的間隔短回文重復(fù)序列(clusteredregularlyinterspacedshortpalindromicrepeats,crispr)。crispr是細菌用來抵御病毒侵襲/躲避哺乳動物免疫反應(yīng)的基因系統(tǒng)。crispr廣泛存在于眾多原核生物基因中,其中ii型為crispr/cas,免疫系統(tǒng)依賴cas9內(nèi)切酶家族靶向和剪切外源dna。在這一系統(tǒng)中,crrna(crispr-derivedrna)通過堿基配對與trancrrna(trans-activatingrna)結(jié)合形成雙鏈rna,此trancrrna/crrna二元復(fù)合體(現(xiàn)在融合成為了sgrna,singleguidedrna)指導(dǎo)cas9蛋白在crrna引導(dǎo)序列靶向位點剪切雙鏈dna,其中cas9的hnh核酸酶結(jié)構(gòu)域剪切互補鏈,其ruvc-like結(jié)構(gòu)域剪切非互補鏈,形成雙鏈dna缺口,然后細胞會借助同源重組機制(homologousrecombination)或者非同源末端連接機制(non-homologousendjoining)對斷裂的dna進行修復(fù)。如果細胞通過非同源重組機制進行修復(fù),將會產(chǎn)生隨機的突變、缺失或插入,造成基因敲除的效果。如果細胞通過同源重組機制進行修復(fù),會用另外一段dna片段填補斷裂的dna缺口,因而會引入一段“新的”遺傳信息,造成基因敲入效果。
技術(shù)實現(xiàn)要素:
本發(fā)明的目的是同時實現(xiàn)無物種限制的真核生物的基因敲除和基因過表達。
根據(jù)本發(fā)明的第一方面,提供了一種(非治療目的)多基因同時過表達和/或敲除方法,包括:
將至少一個car構(gòu)建于同一載體,并用同一個啟動子驅(qū)動表達,并且不同car之間利用2a短肽分開;和/或
將至少一個sgrna構(gòu)建于同一載體,不同sgrna分別用啟動子u6驅(qū)動表達,并且不同的u6-sgrna串聯(lián)在一起;
將所述載體電轉(zhuǎn)染(脂質(zhì)體轉(zhuǎn)染或電穿孔)導(dǎo)入同一細胞或個體,其中所述至少一個car在同一細胞或個體同時實現(xiàn)基因過表達,所述至少一個sgrna在同一細胞或個體同時實現(xiàn)基因敲除。
根據(jù)本發(fā)明的一個具體實施例,所述至少一個sgrna可以在同一細胞或個體同時實現(xiàn)敲除胞內(nèi)表達靶基因和胞膜表達靶基因,或同時實現(xiàn)敲除一條信號通路的上下游靶基因或幾條平行信號通路的不同靶基因。所述至少一個sgrna優(yōu)選為兩個分別靶向胞內(nèi)表達基因hgata3和胞膜表達基因hpd1的sgrna。
根據(jù)本發(fā)明的一個具體實施例,所述至少一個car可以為單個過表達的tn-muc1car或her2car,所述至少一個sgrna可以為單個靶向胞內(nèi)表達靶基因hgata3的sgrna,并且car載體和sgrna載體(的dna片段)被串聯(lián)構(gòu)建為一體。
根據(jù)本發(fā)明的一個具體實施例,所述至少一個car可以為過表達的tn-muc1car和/或her2car,所述至少一個sgrna可以為兩個分別靶向胞內(nèi)表達靶基因hgata3和胞膜表達靶基因hpd1的sgrna,并且car載體和sgrna載體被串聯(lián)構(gòu)建為一體。
上述實施例中,所述細胞優(yōu)選為hek293t細胞或人原代t細胞。
根據(jù)本發(fā)明,tn-muc1car或her2car是優(yōu)選利用piggybac-transposon載體依次串聯(lián)人啟動子、csf2ra嵌合受體信號肽、胞膜外抗原結(jié)合區(qū)、鉸鏈區(qū)、胞內(nèi)信號傳導(dǎo)區(qū)和t2a短肽連接的抗性基因puromycin制備而成的。
胞膜外抗原結(jié)合區(qū)優(yōu)選為用于結(jié)合tn-muc1或her2蛋白的cd19單鏈抗體(scfv),依次串聯(lián)c-myc表位標(biāo)記、cd8hinge嵌合受體鉸鏈、cd8transmembrane嵌合受體跨膜區(qū)。
胞內(nèi)信號傳導(dǎo)區(qū)優(yōu)選為cd28-4-1bb-cd3ζ,cd28和4-1bb為嵌合受體共刺激因子。
根據(jù)本發(fā)明的另一方面,提供了一種多基因同時過表達和/或敲除裝置,包括:
pbmc細胞準(zhǔn)備系統(tǒng);
amaxa電轉(zhuǎn)試劑盒;
電轉(zhuǎn)染系統(tǒng),接收來自amaxa電轉(zhuǎn)試劑盒的電轉(zhuǎn)試劑并將其和用于過表達基因的質(zhì)粒和用于基因敲除的質(zhì)粒混合,形成電轉(zhuǎn)混合物體系;并接收來自pbmc細胞準(zhǔn)備系統(tǒng)的pbmc細胞且在其中加入所述混合物進行電轉(zhuǎn)染;
cd3陽性t細胞富集系統(tǒng),包含用于偶聯(lián)cd3/cd28抗體的磁珠;以及
篩培系統(tǒng),用于篩選并培養(yǎng)擴增轉(zhuǎn)染后的t細胞。
本發(fā)明的又一目的是提供一種根據(jù)上述方法獲得的分離的t細胞或細胞系或其次代培養(yǎng)物。
本發(fā)明選用piggybac轉(zhuǎn)座子系統(tǒng)提高了轉(zhuǎn)染效率和轉(zhuǎn)基因的表達效率,縮短了制備時間,簡化了car-t的制備程序,增強了系統(tǒng)的負載量。另外,本發(fā)明敲除基因時避免了使用病毒載體,安全性更高。
本發(fā)明將基因敲除與car的裝備放在同一個步驟里完成,提高了car-t細胞的制備效率和存活率,降低了多次操作的成本;同時縮短了car-t細胞在體外的培養(yǎng)周期,保證了其在體外擴增的活性。
本發(fā)明通過構(gòu)建一個載體,利用不同的啟動子同時過表達某種基因,實現(xiàn)基因的過表達,同時通過cas9和sgrna實現(xiàn)基因敲除。本發(fā)明因此實現(xiàn)了在同一細胞或同一個體同時過表達基因和敲除基因的目的。另外,本發(fā)明還利用自我剪切的2a短肽(包括p2a、t2a兩種)連接不同基因,保證多基因的等量同時過表達;串聯(lián)多個u6-sgrna(靶向不同靶基因)同時實現(xiàn)多個基因的敲除。本發(fā)明因此實現(xiàn)了在同一細胞或同一個體同時實現(xiàn)多個基因的過表達和多個基因的敲除,特別是可以同時敲除胞內(nèi)表達靶基因和胞膜表達靶基因,或同時敲除一條信號通路的上下游靶基因,或幾條平行信號通路的不同靶基因。
本發(fā)明的再一目的是構(gòu)建同時進行基因敲除和基因過表達的載體。
本發(fā)明通過合理構(gòu)建載體,可一次性簡單實現(xiàn)同一個體或同一細胞同時進行基因敲除和基因過表達,且消耗低、耗時短、效率高,是實現(xiàn)真核生物特別是單細胞基因敲除和基因過表達的引領(lǐng)方法。

具體實施方式
構(gòu)建過表達一個和多個基因的載體
(1)以過表達一種抗tn-muc1嵌合抗原受體(tn-muc1car)為例,與傳統(tǒng)的利用病毒做car載體不同,本發(fā)明采用第三代car,將tn-muc1car整合到piggybac轉(zhuǎn)座子表達系統(tǒng)中。這樣一是回避了病毒載體可能導(dǎo)致的生物安全性問題;二是可以延長car表達的時間;三是制備簡單,不用像病毒包裝一樣,需要非常嚴(yán)格的條件。
圖1展示第三代car的組成元件,包括嵌合受體信號肽、胞膜外抗原結(jié)合區(qū)(scfv)、跨膜區(qū)(cd8αtm)、胞內(nèi)信號傳導(dǎo)區(qū)(cd28,4-1bb,cd3ζ)。圖2展示tn-muc1carpiggybac表達載體中的主要元件,car基因包含有一個經(jīng)過人工優(yōu)化過的信號肽,tn-muc1胞膜外抗原結(jié)合區(qū),c-myc標(biāo)簽,cd8跨膜區(qū),共激活受體cd28和4-1bb胞內(nèi)信號傳導(dǎo)區(qū),cd3ζ胞內(nèi)信號傳導(dǎo)區(qū)。car基因與藥篩抗性基因puromycin通過具有自我剪切功能的2a肽段相連,在ef1α啟動子的驅(qū)動下同時表達。載體的兩端是反向重復(fù)序列(ir)。
載體具體構(gòu)建如下:anti-tn-muc1抗體的scfv序列經(jīng)人工合成嵌合進puc57表達載體中,分別設(shè)計帶有xba1酶切位點的正向引物和帶有bgl2酶切位點的反向引物將scfv序列經(jīng)聚合酶鏈?zhǔn)椒磻?yīng)(pcr)擴增出來。擴增產(chǎn)物與piggybac-cd19car載體同時進行xba1和bgl2酶切,酶切產(chǎn)物經(jīng)純化后連接,轉(zhuǎn)化,構(gòu)建出piggybac-tn-muc1car。載體主要元件序列詳見序列表中的序列1。
(2)再以制備一種抗her2嵌合抗原受體(her2car)為例,同樣采用第三代car,并且將her2car整合進了piggybac轉(zhuǎn)座子表達系統(tǒng)中。載體中的元件與載體tn-muc1car一致(圖3)。
具體構(gòu)建如下:anti-her2抗體的scfv序列經(jīng)人工合成嵌合進puc57表達載體中,分別設(shè)計帶有xba1酶切位點的正向引物和帶有bgl2酶切位點的反向引物將scfv序列經(jīng)聚合酶鏈?zhǔn)椒磻?yīng)(pcr)擴增出來。擴增產(chǎn)物與piggybac-cd19car載體同時進行xba1和bgl2酶切,酶切產(chǎn)物經(jīng)純化后連接,轉(zhuǎn)化,構(gòu)建出piggybac-her2car。載體主要元件序列詳見序列表中的序列2。
(3)再以同時表達兩種car為例,本發(fā)明構(gòu)建一種同時表達由短肽p2a連接起來的tn-muc1car和her2car基因的載體(圖4),兩個car都采用第三代car,并同時整合進piggybac轉(zhuǎn)座子表達載體中。
具體構(gòu)建如下:分別設(shè)計引物將圖2中tn-muc1car和圖3中her2car基因表達序列通過聚合酶鏈?zhǔn)椒磻?yīng)擴增出來,擴增tn-muc1正向引物中包含有20個堿基的同源臂,可以與piggybac載體同源重組,擴增tn-muc1反向引物中包含有p2a序列;擴增her2正向引物中包含有20個堿基的同源臂,可以與tn-muc1中p2a同源重組,擴增her2反向引物中包含有20個可以與t2a同源重組的序列。piggybac-cd19car載體經(jīng)xba1和bamh1雙酶切,酶切產(chǎn)物與擴增出來的tn-muc1片段和her2片段同源重組,構(gòu)建成piggybac-tn-muc1-p2a-her2car。載體主要元件序列詳見序列表中的序列3。
構(gòu)建cas9/sgrna介導(dǎo)單基因敲除載體
(1)以gata3和shp1為例,利用cas9/sgrna介導(dǎo)胞內(nèi)蛋白gata3或者shp1的基因敲除。
cas9表達載體為pst1374-nls-flag-cas9zf-nls(載體主要元件序列詳見序列表中的序列4),載體上帶有藥篩基因blasticidin(bsd)(圖5)。sgrna的表達載體為pgl3-u6-sgrna(載體主要元件序列詳見序列表中的序列5),載體上帶有藥篩基因puromycin(puro)(圖6)。humangata3sgrna載體的構(gòu)建:在gata3基因的外顯子2(e2)中,根據(jù)gn19ngg或者是n20ngg的原則設(shè)計20個堿基的互補配對的上下游引物,經(jīng)過退火之后連接到pgl3-u6-sgrna載體上,構(gòu)成pgl3-u6-hgata3sgrna(圖7)。humanshp1sgrna載體的構(gòu)建:根據(jù)相同的原則,在shp1基因的外顯子3(e3)上設(shè)計20個堿基的guiderna序列,經(jīng)退火連接之后構(gòu)成pgl3-u6-hshp1sgrna(圖8)。
(2)以pd1為例,利用cas9/sgrna介導(dǎo)膜蛋白pd1的基因敲除。cas9表達載體同圖5。pd1sgrna載體的構(gòu)建:在pd1基因的外顯子2(e2)中,根據(jù)gn19ngg或者是n20ngg的原則設(shè)計20個堿基的互補配對的上下游引物,經(jīng)過退火之后連接到pgl3-u6-sgrna載體上,構(gòu)成pgl3-u6-hpd1sgrna(圖9)。
(3)單一cas9/sgrna載體所介導(dǎo)的基因敲除。
cas9/sgrna所介導(dǎo)的基因敲除,必須要在sgrna的介導(dǎo)下,將cas9蛋白牽引到基因組上特定的位點進行基因編輯,因此在細胞上實現(xiàn)單一基因的敲除必須在同一個細胞上同時表達cas9和sgrna,而當(dāng)cas9和sgrna表達在不同載體上時,由于轉(zhuǎn)染感染或者是電轉(zhuǎn)的效率問題,在一個細胞中可能僅表達一個載體,這樣就會限制了基因編輯的效率,因此,將cas9和sgrna構(gòu)建在同一個載體上將會極大的提高基因編輯的效率。如圖10所示,cas9/sgrna共表達載體為pst1374-nls-flag-cas9zf-nls-u6-sgrna(載體主要元件序列詳見序列表中的序列6),cas9蛋白與藥篩基因(puro)在cmv啟動子的驅(qū)動下同時表達,兩者之間以具有自我剪切功能的多肽t2a相連,sgrna在u6啟動子的驅(qū)動下單獨表達。根據(jù)圖7、圖8、圖9所示的序列,設(shè)計20個堿基的互補配對的上下游引物,經(jīng)過退火之后連接到pst1374-nls-flag-cas9zf-nls-u6-sgrna載體上,本發(fā)明構(gòu)建了pst1374-nls-flag-cas9zf-nls-u6-hgata3sgrna,pst1374-nls-flag-cas9zf-nls-u6-hshp1sgrna和pst1374-nls-flag-cas9zf-nls-u6-hpd1sgrna。
構(gòu)建cas9/sgrna介導(dǎo)多基因敲除載體
cas9/sgrna介導(dǎo)的基因編輯的另一個優(yōu)勢是可以同時進行多基因的敲除。設(shè)計分別針對不同靶基因的sgrna,各個sgrna分別由一個u6啟動子啟動表達。將多個u6-sgrna串在一起,通過各個u6的介導(dǎo),實現(xiàn)多個sgrna的表達。在多個sgrna的介導(dǎo)下,可以同時實現(xiàn)多個位點的同時基因編輯,為此,我們構(gòu)建了可以同時敲除胞內(nèi)表達蛋白hgata3和膜表面蛋白hpd1的載體pst1374-nls-flag-cas9zf-nls-u6-hgata3sgrna-u6-hpd1sgrna(圖11)(載體主要元件序列詳見序列表中的序列7)。
構(gòu)建同時過表達基因和sgrna的載體
同時實現(xiàn)基因過表達和基因敲除的最好表達載體是將過表達基因和基因敲除元件構(gòu)建在同一個載體上。實現(xiàn)基因的過表達需要將外源性的基因表達片段整合入細胞的基因組中實現(xiàn)持續(xù)的穩(wěn)定的表達,而cas9/sgrna系統(tǒng)因為存在脫靶效應(yīng),因此不能同時在細胞中穩(wěn)定表達,因此,盡管piggybac載體可以同時表達外源基因片段、cas9蛋白和sgrna,還是應(yīng)當(dāng)盡量避免由cas9/sgrna所帶來的脫靶效應(yīng)。本發(fā)明將外源性的基因與sgrna融合進piggybac表達載體中,cas9蛋白的表達與轉(zhuǎn)座酶基因的表達則構(gòu)建在另一個載體之中。
同時表達cas9蛋白和轉(zhuǎn)座酶的表達載體如圖12所示。在cmv啟動子的驅(qū)動下,cas9蛋白、轉(zhuǎn)座酶和藥篩基因blasticidin同時表達,cas9蛋白和轉(zhuǎn)座酶之間以具有自我剪切功能的多肽p2a相連,轉(zhuǎn)座酶和藥篩基因blasticidin之間以具有自我剪切功能的多肽t2a相連(載體主要元件序列詳見序列表中的序列8)。
(1)為實現(xiàn)同一細胞或個體內(nèi)同時過表達某一基因和敲除某一基因,以car為例,本發(fā)明構(gòu)建了同時表達抗tn-muc1嵌合抗原受體和humangata3sgrna的載體(載體主要元件序列詳見序列表中的序列9),如圖13所示??箃n-muc1嵌合抗原受體與藥篩基因(puro)在啟動子ef1的啟動下同時表達,兩者之間以具有自我剪切功能的多肽t2a相連接。humangata3sgrna在u6啟動子的驅(qū)動下表達。
(2)以car和sgrna為例,按照相同的策略,本發(fā)明構(gòu)建了同時表達抗tn-muc1嵌合抗原受體和humanshp1sgrna的載體。
(3)以car和sgrna為例,按照相同的策略,本發(fā)明構(gòu)建了同時表達抗tn-muc1嵌合抗原受體和humangata3sgrna以及humanpd1sgrna的載體(載體主要元件序列詳見序列表中的序列10)(圖14)。
在常規(guī)細胞上同時實現(xiàn)基因過表達和單基因敲除
本發(fā)明采用兩個質(zhì)粒轉(zhuǎn)染法(以下轉(zhuǎn)染都采用兩質(zhì)粒轉(zhuǎn)染法),利用脂質(zhì)體或電轉(zhuǎn)將表達tn-muc1-humangata3sgrna的載體與表達cas9-p2a-transposase的載體一起轉(zhuǎn)染至hek293t細胞中,以lipo2000為例,具體步驟如下:
1、真核生物細胞的培養(yǎng)與轉(zhuǎn)染
(1)以hek293t細胞為例:hek293t細胞接種培養(yǎng)于添加10%fbs的dmem高糖培養(yǎng)液中(hyclone,sh30022.01b),其中含penicillin(100u/ml)和streptomycin(100μg/ml)。
(2)在轉(zhuǎn)染前分至6孔板中,待密度達到70%-80%時進行轉(zhuǎn)染。
(3)按照lipofectaminetm2000transfectionreagent(invitrogen,11668-019)的操作手冊,將2μgpiggybac-ef1-tn-muc1-u6-hgata3質(zhì)粒與2μg的pst1374-nls-flag-cas9zf-nls-p2a-transposase質(zhì)?;靹?共轉(zhuǎn)染至每孔細胞中,6-8小時后換液,12小時后,向每孔細胞中加入10μg/mlblasticidin(sigma,15205)和2μg/mlpuromycin(merck,540411)藥篩,72小時后收取細胞。
2、基因敲除的檢測(以t7en1酶切為例)
(1)收取部分細胞在裂解液(10μmtris-hcl,0.4mnacl,2μmedta,1%sds)中用100μg/ml蛋白酶k裂解消化后,酚-氯仿抽提后溶解到50μl去離子水中。
(2)使用一對引物hgata3t7en1for和hgata3t7en1rev進行pcr擴增,用axypreppcrcleanup純化獲得pcr回收產(chǎn)物,取200ng統(tǒng)一稀釋到20μl進行變性、退火,程序如:95℃,5min;95–85℃at-2℃/s;85–25℃at-0.1℃/s;holdat4℃。
(3)在20μl體系中加入t7nei內(nèi)切酶(t7en1)0.3μl,37℃酶切30分鐘后,加入2μl10xloadingbuffer,用3%的瓊脂糖膠電泳檢測,結(jié)果見圖15a。
3、ta克隆測序
(1)將t7en1酶切檢測步驟(2)獲得的pcr回收產(chǎn)物用rtaq進行加a反應(yīng)。加a反應(yīng)體系為:
700~800ngpcr回收產(chǎn)物
5μl10xbuffer(mg2+plus)
4μldntp
0.5μlrtaq(takara,r001am)
補水至50μl體系。
37℃溫育30分鐘后,取1μl產(chǎn)物與pmd19-tvector(takara,3271)連接并轉(zhuǎn)化dh5感受態(tài)細胞(transgen,cd201)。
(2)挑取單克隆,用通用引物m13-f測序,測序結(jié)果見圖15b,結(jié)果發(fā)現(xiàn):靶基因hgata3缺失了sgrna靶向的一段序列,基因敲除成功。
4、真核生物表達外源基因的檢測(以tn-muc1為例)
(1)收取細胞,在pbs中清洗一次,1000轉(zhuǎn),5分鐘離心,棄掉上清。
(2)一抗染色:mouseanti-c-myc抗體(cellsignaling,2276s)1:500稀釋,將細胞重懸在100μl體系中,冰上放置10分鐘。
(3)終止反應(yīng):向體系中加入1mlpbs終止染色反應(yīng),1000轉(zhuǎn),離心5分鐘,棄掉上清。
(4)二抗染色:goatanti-mousepe抗體1:10稀釋,將細胞重懸在100μl體系中,冰上放置10分鐘。
(5)終止反應(yīng):向體系中加入1mlpbs終止染色反應(yīng),1000轉(zhuǎn),離心5分鐘,棄掉上清。
(6)以500μlpbs重懸細胞,流式檢測。流式檢測結(jié)果見圖15c。
以人的t細胞為例,本發(fā)明在原代細胞同時實現(xiàn)基因的過表達和單基因敲除
1、pbmc細胞的分離純化:
(1)用抗凝管采集外周血,邊采集邊搖晃使外周血與抗凝劑充分混合;
(2)外周血細胞與淋巴細胞分離液等體積混合,離心,吸取離心后的白膜層細胞;
(3)將得到的白膜層細胞與pbs或者無血清細胞培養(yǎng)基1640混合后離心,沉淀即為所述pbmc細胞。
重復(fù)三遍。
2、cd3陽性細胞的富集
(1)調(diào)整pbmc細胞濃度至50x106cell/ml。
(2)按每1ml加入cd3+enrichedantibodiescocktail50μl,混勻后室溫靜置5分鐘。
(3)按每1ml加入magnet150μl,混勻后室溫靜置10分鐘,
(4)將離心管置于磁力架上靜置5分鐘,吸取上層細胞懸液至新的15ml離心管中。
(5)重復(fù)該操作一次。
(6)室溫離心300*g,10分鐘,收集細胞。
(7)細胞計數(shù)。
3、cd3陽性細胞的電轉(zhuǎn)
(1)配置電轉(zhuǎn)體系
向1.5ml離心管中分別加入8μgpiggybac-ef1a-tn-muc1-u6-hgata3質(zhì)粒和8μg的pst1374-nls-flag-cas9zf-nls-p2a-transposase質(zhì)粒,并按照lonzaamaxa電轉(zhuǎn)試劑盒說明書要求,加入82μl電轉(zhuǎn)緩沖液和18μlsupplement1,混勻。對照組中僅過表達tn-muc1而不進行基因敲除。
(2)收取20x106個細胞到15ml離心管中,300g離心10分鐘,棄掉上清。
(3)以(1)中配好的質(zhì)粒電轉(zhuǎn)緩沖液混合物重懸細胞,并轉(zhuǎn)移至電轉(zhuǎn)杯中。
(4)使用儀器lonza2b,u-014程序進行電轉(zhuǎn)。
(5)電轉(zhuǎn)后的細胞迅速轉(zhuǎn)移至提前預(yù)熱的添加有10%fbs的aim-v培養(yǎng)基中,37度5%二氧化碳培養(yǎng)箱中培養(yǎng)2小時。
(6)電轉(zhuǎn)后的細胞全換液,以1x106個/ml的密度重懸細胞,培養(yǎng)過夜。
(7)24小時后,取1x105個細胞用流式檢測電轉(zhuǎn)效率。
4、t細胞的激活培養(yǎng)
(1)電轉(zhuǎn)培養(yǎng)24小時后,向培養(yǎng)基中加入100u/mlil-2,并按照1:1的比例加入cd3/cd28dynabeads,激活t細胞。
(2)每兩天對細胞半換液,或者是補加il-2,細胞密度始終維持在1x106個/ml。
(3)激活5天后,將t細胞收集到15ml離心管中,并將離心管置于磁力架中,慢慢將上清轉(zhuǎn)移到另外一個干凈的15ml離心管中,重復(fù)此步驟一次。
(4)室溫離心300*g,10分鐘,棄上清,使用10%fbs,300u/mlil-2aim-v培養(yǎng)基重懸細胞,密度控制在1x106個/ml。
(5)每兩天對細胞半換液,或者是補加il-2,并計數(shù),細胞密度始終維持在1x106個/ml。
(6)培養(yǎng)到第14天的時候,計數(shù),取1x106個細胞流式檢測tn-muc1car的表達并進行t7en1檢測hgata3基因的敲除效率。
5、基因敲除以及外源基因表達的檢測
檢測方法請參照普通細胞檢測的方法,t7en1的檢測結(jié)果詳見圖16a,ta克隆的結(jié)果詳見圖16b。流式檢測tn-muc1car的表達結(jié)果見圖16c。
在常規(guī)細胞上同時實現(xiàn)基因過表達和多基因敲除
以hek293t細胞為例,本發(fā)明在真核生物細胞同時實現(xiàn)基因的過表達和多基因敲除。本發(fā)明同時在hek293t細胞上敲除胞內(nèi)蛋白gata3和細胞膜表面蛋白pd1,并同時過表達tn-muc1car基因。具體操作步驟請參照在常規(guī)細胞上同時實現(xiàn)基因過表達和單基因敲除的具體操作步驟。
