本發(fā)明涉及一種鑒別蠶絲種類的方法,更具體的說是涉及一種鑒別古代絲織品蠶絲種類的方法。
背景技術(shù):
蠶絲是古代漢族文明產(chǎn)物之一,是熟蠶結(jié)繭時分泌絲液凝固而成的連續(xù)長纖維,也稱“天然絲”,根據(jù)食物的不同,又分桑蠶、柞蠶、木薯蠶、樟蠶、柳蠶和天蠶等,各種蠶絲在生活領(lǐng)域應(yīng)用廣泛,在學(xué)術(shù)領(lǐng)域也備受關(guān)注,但是由不同蠶絲制成的各種絲織品,往往會因為工作的不慎或者時間的延長而導(dǎo)致標簽?zāi):煜烁鞣N絲織品,給之后的科研工作帶來很大的不便,如果是保存較好的絲織品可以通過紅外,XRD等多種方式加以區(qū)分其蠶絲種類,對于保存狀態(tài)不好的古代絲織品使用普通的分析測試技術(shù)則很難鑒別。
技術(shù)實現(xiàn)要素:
本發(fā)明的目的在于克服現(xiàn)有技術(shù)存在的不足,而提供一種快速,直觀,簡便的鑒別古代絲織品蠶絲種類的方法。
為實現(xiàn)上述目的,本發(fā)明提供了如下技術(shù)方案:一種鑒別古代絲織品蠶絲種類的方法,該方法采用如下步驟:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各2~8g,分為A,B,C三組;
b)三組樣品均以1:100的浴比在質(zhì)量分數(shù)為0.4~0.6%的Na2CO3水溶液中沸煮20~40min進行脫膠,反復(fù)4~6次,然后將三組樣品溶液分別和絲素分離,待絲素用去離子水沖洗絞干后,再取50~80ml質(zhì)量分數(shù)為1%的鹽酸溶液,在室溫下浸放50~60min,用溫水洗至溶液呈中性,在60~80℃的烘箱中烘干;
c)將由步驟b)脫鹽烘干的絲素繼續(xù)脫色和脫油蠟質(zhì),先在50~70℃溫度下,放入90~110ml的質(zhì)量分數(shù)為60~80%甲醇溶液中浸100~120min,再在50~70℃溫度下,放入90~110ml無水乙醇中浸60~80min,接著在30~50℃溫度下,放入90~110ml的丙酮溶液中浸60~80min,最后絞干浸入90~110ml的乙醚中50~70min,使用吹風機吹干;
d)將由步驟c)得到的絲素在96~98℃下煮50~60min,反復(fù)4~6次,在60~80℃的烘箱中烘干;
e)由19.8g氫氧化銅和25.5ml乙二胺配制300ml的銅乙二胺溶液,將由步驟d)處理之后的三組絲素樣品分別放入配好的銅乙二胺溶液中,浴比為1:50,在50~60℃下保溫1~2h;
f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至8~10,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的1/3~2/3;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為1.5~2%的二硫蘇糖醇溶液2~3ml于40~50℃下保持15~20min,待冷卻至室溫加入質(zhì)量分數(shù)為2.2~2.8%的碘乙酰胺溶液4~5ml,黑暗環(huán)境下保持15~20min后加入50~80ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;
h)采用HPLC-MS/MS對其進行質(zhì)譜分析,將A,B,C三組實驗的氨基酸序列與氨基酸序列數(shù)據(jù)庫進行對比,根據(jù)特征肽段即可確定A,B,C三組絲織品蠶絲的種類。
一種鑒別古代絲織品蠶絲種類的方法,其特征在于采用步驟如下:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各5g,分為A,B,C三組;
b)三組樣品均以1:100的浴比在質(zhì)量分數(shù)為0.5%的Na2CO3水溶液中沸煮30min進行脫膠,反復(fù)5次,然后將三組樣品溶液分別和絲素分離,待絲素用去離子水沖洗絞干后,再取65ml質(zhì)量分數(shù)為1%的鹽酸溶液,在室溫下浸放55min,用溫水洗至溶液呈中性,在70℃的烘箱中烘干;
c)將由步驟b)脫鹽烘干的絲素繼續(xù)脫色和脫油蠟質(zhì),先在60℃溫度下,放入100ml的質(zhì)量分數(shù)為70%甲醇溶液中浸110min,再在60℃溫度下,放入100ml無水乙醇中浸70min,接著在40℃溫度下,放入100ml的丙酮溶液中浸70min,最后絞干浸入100ml的乙醚中60min,使用吹風機吹干;
d)將由步驟c)得到的絲素在97℃下煮55min,反復(fù)5次,在70℃的烘箱中烘干;
e)由19.8g氫氧化銅和25.5ml乙二胺配制300ml的銅乙二胺溶液,將由步驟d)處理之后的三組絲素樣品分別放入配好的銅乙二胺溶液中,浴比為1:50,在55℃下保溫1.5h;
f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至9,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的1/2;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為1.7%的二硫蘇糖醇溶液2.5ml于45℃下保持17min,待冷卻至室溫加入質(zhì)量分數(shù)為2.5%的碘乙酰胺溶液4.5ml,黑暗環(huán)境下保持17min后加入65ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;
h)采用HPLC-MS/MS對其進行質(zhì)譜分析,將A,B,C三組實驗的氨基酸序列與氨基酸序列數(shù)據(jù)庫進行對比,根據(jù)特征肽段即可確定A,B,C三組絲織品蠶絲的種類。
本發(fā)明的有益效果是:采用蛋白質(zhì)組學(xué)方法鑒別古代絲織品蠶絲種類,克服了普通檢測分析方法的檢測限度,使得即便是保存狀態(tài)不好的古代絲織品也可以被區(qū)分鑒定;采用蛋白質(zhì)組學(xué)方法鑒定古代絲織品蠶絲種類,檢測中使用溶液內(nèi)酶切的技術(shù)使得實驗操作簡單方便;采用蛋白質(zhì)組學(xué)方法鑒定古代絲織品蠶絲種類,靈敏度高,準確度高,樣品使用量少。
具體實施方式
以下結(jié)合具體實施例對本發(fā)明作詳細介紹:
一種鑒別古代絲織品蠶絲種類的方法,該方法采用如下步驟:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各2~8g,分為A,B,C三組;
b)三組樣品均以1:100的浴比在質(zhì)量分數(shù)為0.4~0.6%的Na2CO3水溶液中沸煮20~40min進行脫膠,反復(fù)4~6次,然后將三組樣品溶液分別和絲素分離,待絲素用去離子水沖洗絞干后,再取50~80ml質(zhì)量分數(shù)為1%的鹽酸溶液,在室溫下浸放50~60min,用溫水洗至溶液呈中性,在60~80℃的烘箱中烘干;

d)將由步驟c)得到的絲素在96~98℃下煮50~60min,反復(fù)4~6次,在60~80℃的烘箱中烘干;
e)由19.8g氫氧化銅和25.5ml乙二胺配制300ml的銅乙二胺溶液,將由步驟d)處理之后的三組絲素樣品分別放入配好的銅乙二胺溶液中,浴比為1:50,在50~60℃下保溫1~2h;
f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至8~10,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的1/3~2/3;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為1.5~2%的二硫蘇糖醇溶液2~3ml于40~50℃下保持15~20min,待冷卻至室溫加入質(zhì)量分數(shù)為2.2~2.8%的碘乙酰胺溶液4~5ml,黑暗環(huán)境下保持15~20min后加入50~80ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;
h)采用HPLC-MS/MS對其進行質(zhì)譜分析,將A,B,C三組實驗的氨基酸序列與氨基酸序列數(shù)據(jù)庫(見附件1)進行對比,根據(jù)特征肽段即可確定A,B,C三組絲織品蠶絲的種類。
實施例1:一種鑒別古代絲織品蠶絲種類的方法,該方法采用如下步驟:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各2g,分為A,B,C三組;

f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至8,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的1/3;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為1.5%的二硫蘇糖醇溶液2ml于40℃下保持15min,待冷卻至室溫加入質(zhì)量分數(shù)為2.2%的碘乙酰胺溶液4ml,黑暗環(huán)境下保持15min后加入50ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;

實施例2:一種鑒別古代絲織品蠶絲種類的方法,該方法采用如下步驟:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各5g,分為A,B,C三組;
b)三組樣品均以1:100的浴比在質(zhì)量分數(shù)為0.5%的Na2CO3水溶液中沸煮30min進行脫膠,反復(fù)5次,然后將三組樣品溶液分別和絲素分離,待絲素用去離子水沖洗絞干后,再取65ml質(zhì)量分數(shù)為1%的鹽酸溶液,在室溫下浸放55min,用溫水洗至溶液呈中性,在70℃的烘箱中烘干;
c)將由步驟b)脫鹽烘干的絲素繼續(xù)脫色和脫油蠟質(zhì),先在60℃溫度下,放入100ml的質(zhì)量分數(shù)為70%甲醇溶液中浸110min,再在60℃溫度下,放入100ml無水乙醇中浸70min,接著在40℃溫度下,放入100ml的丙酮溶液中浸70min,最后絞干浸入100ml的乙醚中60min,使用吹風機吹干;
d)將由步驟c)得到的絲素在97℃下煮55min,反復(fù)5次,在70℃的烘箱中烘干;
e)由19.8g氫氧化銅和25.5ml乙二胺配制300ml的銅乙二胺溶液,將由步驟d)處理之后的三組絲素樣品分別放入配好的銅乙二胺溶液中,浴比為1:50,在55℃下保溫1.5h;
f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至9,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的1/2;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為1.7%的二硫蘇糖醇溶液2.5ml于45℃下保持17min,待冷卻至室溫加入質(zhì)量分數(shù)為2.5%的碘乙酰胺溶液4.5ml,黑暗環(huán)境下保持17min后加入65ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;
h)采用HPLC-MS/MS對其進行質(zhì)譜分析,將A,B,C三組實驗的氨基酸序列與氨基酸序列數(shù)據(jù)庫進行對比,根據(jù)特征肽段即可確定A,B,C三組絲織品蠶絲的種類。
實施例3:一種鑒別古代絲織品蠶絲種類的方法,該方法采用如下步驟:
a)分別取一份已被混淆的由桑蠶絲,柞蠶絲,栗蠶絲制作的古代絲織品各8g,分為A,B,C三組;
b)三組樣品均以1:100的浴比在質(zhì)量分數(shù)為0.6%的Na2CO3水溶液中沸煮40min進行脫膠,反復(fù)6次,然后將三組樣品溶液分別和絲素分離,待絲素用去離子水沖洗絞干后,再取80ml質(zhì)量分數(shù)為1%的鹽酸溶液,在室溫下浸放60min,用溫水洗至溶液呈中性,在80℃的烘箱中烘干;
c)將由步驟b)脫鹽烘干的絲素繼續(xù)脫色和脫油蠟質(zhì),先在70℃溫度下,放入110ml的質(zhì)量分數(shù)為80%甲醇溶液中浸120min,再在70℃溫度下,放入110ml無水乙醇中浸80min,接著在50℃溫度下,放入110ml的丙酮溶液中浸80min,最后絞干浸入110ml的乙醚中70min,使用吹風機吹干;
d)將由步驟c)得到的絲素在98℃下煮60min,反復(fù)6次,在80℃的烘箱中烘干;
e)由19.8g氫氧化銅和25.5ml乙二胺配制300ml的銅乙二胺溶液,將由步驟d)處理之后的三組絲素樣品分別放入配好的銅乙二胺溶液中,浴比為1:50,在60℃下保溫2h;
f)待冷卻后使用質(zhì)量分數(shù)為10%的醋酸調(diào)節(jié)pH值至10,采用透析法和超濾法共用對樣品進行脫鹽處理,然后使用聚乙二醇6000濃縮至原體積的2/3;
g)分別取一份三組樣品濃縮之后的溶液,均加入質(zhì)量分數(shù)為2%的二硫蘇糖醇溶液3ml于50℃下保持20min,待冷卻至室溫加入質(zhì)量分數(shù)為2.8%的碘乙酰胺溶液5ml,黑暗環(huán)境下保持20min后加入80ul的胰凝乳蛋白酶溶液然后用甲酸溶液稀釋;
h)采用HPLC-MS/MS對其進行質(zhì)譜分析,將A,B,C三組實驗的氨基酸序列與氨基酸序列數(shù)據(jù)庫進行對比,根據(jù)特征肽段即可確定A,B,C三組絲織品蠶絲的種類。
